Resumo

O metotrexato é uma droga antineoplásica que age alterando a síntese do DNA e interferindo, portanto, na proliferação celular. O objetivo deste trabalho foi verificar o efeito do metotrexato sobre o reparo ósseo após condilectomia unilateral em camundongos. Foram utilizados 31 camundongos (Mus musculus), albinos, de ambos os gêneros, com 30 dias de idade. Esses animais foram anestesiados com 0,5 mL/100 g de peso com hidrato de cloral a 10% por via intraperitoneal e submetidos à condilectomia unilateral, sendo distribuídos em dois grupos. Os animais do Grupo I receberam uma injeção de 15 mg/kg de peso por via intraperitoneal de metotrexato logo após a cirurgia. Os animais do Grupo II receberam uma injeção de soro fisiológico, em volume correspondente ao administrado aos animais do Grupo I, por via intraperitoneal. Sacrificados 5, 10, 15, 30 e 45 dias após as cirurgias, tiveram suas cabeças fixadas, descalcificadas e processadas para inclusão em parafina. A evolução do processo de reparo mostrou uma reorganização crescente das estruturas, culminando por restaurar as características de uma articulação com todos os seus componentes semelhantes quando comparados os Grupos I e II. Assim, pôde-se concluir que, nas condições utilizadas para a realização deste trabalho, o metotrexato não interferiu no processo de reparo após condilectomia unilateral em camundongos.

Abstract

The methotrexate, an antineoplasic drug, acts altering the DNA synthesis, and thus, interfere on cellular proliferation. The aim of this study was to verify the effect of the methotrexate on bone repair after unilateral condylectomy in mice..There were used 31 mice (Mus musculus), albinus, of both sex, with 30 days of age. The animals were anesthetized with intraperitoneal injection of 0.5 mL/100 g of body weight of a 10% solution of chloral hydrate and submitted to unilateral condylectomy. The animals were separated in two groups. Those of group I received a dose of 15 mg/ kg of body weight of methotrexate through intraperitoneal route soon after the surgery. The animals of the group II received a dose of physiologic serum in a similar volume as that administered to the animals of the group I, also through intraperitoneal route. The animals were sacrificed 5, 10, 15, 30 and 45 days after the surgery. The head of the animals were fixed, decalcified and embedded in paraffin. The evolution of the repair showed comparable between the groups I and II as well as that description found in literature. Thus, we can conclude that, in the conditions used to perform this investigation, the methotrexate did not alter the repair process after unilateral condylectomy in mice.
