Resumo

Estudou-se o efeito tóxico e teratogênico do IPT em uma linhagem não-isogênica de camundongos albinos Mus musculus, com especial referência ao desenvolvimento do palato secundário. O tratamento foi feito utilizando-se a dose final de 8 mg/animal, administrada em épocas diferentes do desenvolvimento fetal e de duas formas diferentes. Os resultados mostraram que o efeito da droga, tanto no desenvolvimento do palato secundário quanto nas demais estruturas embrionárias estudadas, está relacionado à idade fetal na época do tratamento bem como à frequência diária de administração da droga, e não com a dose final utilizada. Por outro lado, tal efeito é reversível com a supressão do tratamento. O efeito embriotóxico é diretamente proporcional à dose diária administrada. Embora ambos os efeitos possam se manifestar independentemente, há indicação de que dependem dos genótipos dos animais utilizados. As alterações observadas parecem ser consequência de perturbações na fisiologia materna e não de um efeito primário do fármaco utilizado.

Abstract

The toxic and teratological effects of IPT on a non isogenic breeding of albinus mice Mus musculus were studied, with special reference to the secondary palate developrnent. The treatment was done using a final dose of 8 mg/animal administered at different times of fetal development and in two different ways. The results showed that the secondary palate development as well as the other embryonic structures studied are related to the fetal age at the treatment period as well as the dally frequence of the drug administration but not with the final dose used. On the other hand, such effects are reversible with the treatment suppression. The embryotoxic effect is in direct ratio to the daily administered dose. ln spite of that both effects could appear independently each other, there is an indication that they are dependent of the genotype of the animais. The changes observed seem to be derived fram disturbs of maternal physiology and not as a primary effect of the drug used.
